Red staining of acid fast bacteria
SPECIMEN REQUIREMENTS
Fixation: Tissue: Any well fixed tissue, however, it is suggested to use formalin fixed tissue.
Sections: Cut Paraffin sections at 5 microns.
COMPONENTS AND REAGENTS
1. Fuchsin Stain Solution - 8oz / 240ml
2. Phenol Dilute Solution - 8oz / 240ml
3. Decolorizing Solution - 8oz / 240ml
4. Methylene Blue Solution - 8oz / 240ml
5. Control Slides 1 Stained and 3 unstained
6. Pap Jars
* Working Carbol Fuchsin Stain
Add 1 part Fuchsin Stain Solution to 6 parts Phenol Solution. Mix . Filtration is necessary.
STAINING PROCEDURE
1. Deparaffinize sections and hydrate to deionized water.
2. Stain sections in * Working Carbol Fuchsin Stain for 20 minutes.
3. Wash sections under tap for water for 5 minutes.
4. Differentiate sections in Decolorization Solution until sections appear pale pink.
5. Wash under tap water two minutes.
6. Counterstain in Methylene Blue Solutions for 2 to 3 seconds.
7. Dehydrate through three changes of absolute alcohol.
8. Clear through two changes of Xylene or Xylene Substitute.

Background - Blue
NOTES
1. Working Carbol Fuchsin Stain has a shelf life of 12 months. Some filtration may have to be performed due to air and temperature during shelf life.
