Acid Mucopolysaccharides
SPECIMEN REQUIREMENTS
Fixation: Tissue: Any well fixed tissue , however, it is suggested to use buffered formalin fixed tissue.
Sections: Cut Paraffin sections 6 microns.
COMPONENTS AND REAGENTS
1. Stock Colloidal Iron Solution - 4oz / 120ml
2. Stock Hydrochloric Acid Solution - 4oz / 120ml
3. Stock Potassium Ferrocyanide Solution - 4oz / 120ml
4. Van Gieson Solution - 4oz / 120ml
5. 1 Stained and 3 unstained control slides
12% Glacial Acetic Acid
Add 12ml of Acetic Acid, Glacial (Reagent) to 88ml of Distilled Water. Mix. Shelf life is 1 year.
Colloidal Iron (Working)
Add 15 ml of Distilled water to 20 ml Stock Colloidal Iron Solution. Mix. Add 5 ml of Acetic Acid , Glacial. Mix.
* PFHAS (Potassium Ferrocyanide/Hydrochloric Acid Solution)
Combine equal parts of Stock Potassium Ferrocyanide Solution with Stock Hydrochloric Acid Solution.
STAINING PROCEDURE
1. Deparaffinize sections and hydrate sections to distilled water.
2. Treat sections in 12% Acetic Acid Solution for 3 min.
3. Stain in Colloidal Iron Working for 1 hour.
4. Rinse sections in two changes of 12% Acetic Acid Solution for 4 minutes in each change.
5. Treat sections in *PFHAS for 20 minutes
6. Wash sections in one change of Tap water for 2 minutes followed by a 2 minute rinse in 1 change of deionized water.
7. Counterstain section in Van Gieson Solution for 7 minutes.
8. Dehydrate sections as usual.
9. Mount using Clearslip.

Acidic Mucopolysaccharides-Blue
Background-Red to Orange
NOTES
1. Distilled Water is recommended to be used on all the procedure steps. Tap water may effect staining results. Always use Distilled Water when rinsing glassware, pipettes, or mixing rods.
2. Working stock solutions should always be made fresh before use.
