Collagenous fibrils, Cartilage, Mucin, Amyloid, and Elastic fibrils
SPECIMEN REQUIREMENTS
Fixation: Zenkers fixative with Mercuric Chloride.
Sections: Cut Paraffin sections at 5 microns.
COMPONENTS AND REAGENTS
1. Acid Fuchsin Stain - 4 oz / 120 ml
2. AnilineBlue/Orange G Stain Solution - 4 oz / 120 ml
3. Phosphomolybdic Acid Solution - 4 oz / 120 ml
4. One stained and three unstained slides
5. Pap Jars
*Weigerts Iodine Solution (Available from IMEB) S7035
5% Sodium Thiosulate
Add 5 gm of Sodium Thiosulfate to 100 ml of distilled water.
STAINING PROCEDURE
1. Deparaffinize sections and hydrate to deionized water.
2. Treat sections in *Weigerts Iodine Solution for 5 minutes to remove mercury pigments.
3. Treat sections in 5% Sodium Thiosulfate for 3 minutes to remove Iodine.
4. Stain section in Acid Fuchsin Solution for 5 minutes.
5. Mordant section in Phosphotungstic Acid for 15 minutes.
6. Rinse sections in deionized or distilled water.
7. Stain section in Aniline Blue/ Orange G for 60 minutes.
8. Dehydrate section through alcohols.
9. Mount slide with Clearslip.
RESULTS
Nuclei Red
Collagenous Fibrils Blue
Erythrocytes Yellow
Elastic Fibers Pale pink to pale yellow
